Reporter and Control Vectors

This repository contains well-characterized reporter constructs and controls for verifying transduction efficiency, measuring expression, and troubleshooting delivery methods.

What's included

Fluorescent Reporters

AAV-compatible:

  • pAAV-CAG-GFP - Green fluorescent protein with strong constitutive CAG promoter
  • pAAV-CAG-tdTomato - Red fluorescent protein, codon-optimized for mammalian expression
  • pAAV-hSyn-EGFP - Neuron-specific expression with human synapsin promoter
  • pAAV-hSyn-DIO-mCherry - Cre-dependent red reporter for conditional expression

Lentiviral:

  • pLenti CMV GFP Puro - Third-generation lentiviral vector with puromycin selection

Plasmid:

  • pcDNA3-EGFP - Mammalian expression plasmid for transfection

CRISPR with reporter:

  • pSpCas9(BB)-2A-GFP (PX458) - CRISPR/Cas9 with GFP co-expression for tracking edited cells

Luciferase Reporters

Quantitative bioluminescent readouts for sensitive expression analysis:

  • pAAV-CAG-FLuc - AAV-compatible firefly luciferase with CAG promoter
  • Luciferase-pcDNA3 - Mammalian expression vector for firefly luciferase
  • pGL4.18 CMV-Luc - Optimized firefly luciferase with CMV promoter

Common applications

  • Test AAV serotype tropism - Use reporters to determine which serotype works best in your cell type before ordering custom AAV
  • Optimize transfection conditions - Screen reagents, cell densities, and timing with fluorescent reporters
  • Validate CRISPR delivery - PX458 combines Cas9 with GFP so you can track which cells received the editing machinery
  • Dual-reporter assays - Combine firefly luciferase with Renilla for normalized measurements

Vector backbone variety

These constructs cover multiple delivery approaches:

  • AAV transfer plasmids - ITR-flanked for AAV packaging (CAG-GFP, CAG-FLuc, hSyn-EGFP)
  • Lentiviral vectors - For stable genomic integration (pLenti CMV GFP Puro)
  • Standard plasmids - For transient transfection (pcDNA3-EGFP, Luciferase-pcDNA3)

Promoter options

  • CAG - Strong constitutive expression in most cell types
